Technical background
Small peptides/haptens must be coupled to carrier proteins (KLH, BSA, Ova) before antibodies can be obtained. In general, polypeptides can be coupled to proteins under the following conditions: 1. There is a free amino or carboxyl group. 2. -SH on cysteine can also be coupled with carrier proteins. At present, our company provides high quality coupled carrier proteins (KLH, BSA, OVA).
Polypeptide antigens are too small to produce a significant immune response, so they need to be coupled to larger protein carriers such as KLH (Keyhole Limpet Hemocyanin), BSA (Bovine Serum Albumin), OVA (Ovalbumin). KLH has no inhibitory effect in ELISA or Western blotting detection, so it does not affect the detection results. The commonly used coupling principle is maleimide theory, which is the coupling of cysteine residues in polypeptides with carrier proteins. Therefore, the addition of cysteine residues in the N-terminal or C-terminal of antigen polypeptide is conducive to the coupling of polypeptide and carrier protein.
Technical principle
In general, polypeptides can be coupled to proteins under the following conditions:
(1) The sulfhydryl group (-SH) on Cys can be coupled (recommended)
(2) or have an unprotected (free) amino or carboxyl group
Peptide KLH coupling modification:
KLH is coupled to a polypeptide by n-terminal NH2
KLH is coupled to polypeptides by C-terminal -COOH
KLH is coupled to a thiol peptide by Cys
Polypeptide BSA coupling modification:
BSA is coupled to a polypeptide by n-terminal NH2
BSA is coupled to polypeptides by C-terminal -COOH
BSA is coupled to Cys by sulfhydryl polypeptides
Polypeptide OVA coupling modification:
The OVA is coupled to the polypeptide by the n-terminal NH2
OVA is coupled to polypeptides by C-terminal -COOH
OVA is coupled to Cys by sulfhydryl polypeptides
